Test opener and Lions captain Alastair Cook returned after missing the three previous matches with a back problem.
The Essex left-hander, looking to prove his fitness ahead of the first Test between England and Pakistan in three weeks' time, faced just three balls before falling lbw to 18-year-old Indian quick Jadie Unadkat.
Cook has only one four-day County Championship match against Yorkshire at Chelmsford before the first Test at Trent Bridge starts on July 29.
